First question: Kinda depends on your individual compute setup and how large your mod list is. Many folks can load their mod list and start the game without issues. But others have found they get better mileage if they, start the game with a smaller list and then after they exit vault 111 wait a couple minutes in-game, save & exit & activate the other mods. ALWAYS start the game with the main game files and patchs, also iirc it's best if frameworks are active from the get go too....

Think the strongest recommendations have been to go-ahead and load most everything as normal at the start, except for the SS2 additional add-on mods (extra plots and such)... load those before you start building anything, but after you've been out of the vault for a bit.
Also, since your starting a fresh game.... make sure you DO NOT have any scraping mods, your game with be a lot better off without them. (they can not be safely removed once a game has been started). These two are safe and recommended to use:
