The tree of choices in this quest seems not to be working quite right when I enter the Hotel Room for the first time.
a) If I ignore the terminal and go straight over to the dresser, I can see Audra's Stash, and then choose to leave it, or spike it with Psycho or Addictol. However at this point I don't have any reason to do anything other than guessing.
b) When I go to the terminal and open the page "Trials", the screen is blank but the quest updates to the next step. Going back in the menu and then re-opening "Trials" then shows me the full page of text which explains the reason why I might want to spike Audra's stash with Addictol (based on Knox's concerns). But now, when I go to the dresser to take action, I can't even "see" Audra's stash at all to make any choices. (I mean, I can see the stack of inhalers, but no choices pop up when I move my mouse pointer over it.) Whether I go to the terminal first or second, in either case after reading the terminal, I can no longer see or spike Audra's stash.
c) If I go downstairs, whether I follow the Knox route or the Anton route, in either case, when I come back upstairs to check on Audra, she is collapsed and incoherent next to the stash, whether or not I spiked the stash earlier with Addictol.

(SS2: character started on 1.0.0f, currently 1.0.0g; SS2E 1.0.0; WF 2.0.5)
 
I tried this again and this time got a result that I can live with:

a) went to dresser first, spiked stash with Addictol. Popup message confirmed I had spiked the stash.
b) went to terminal next, read all pages, the quest updated and told me to check downstairs.
c) sided with Knox, and this time killed Anton myself (before this he had only been wounded); once he he was dead, the quest updated again and Knox prompted me to go back to hotel room
d) we found Audra collapsed next to the stash, but this time she was coherent; she told me I did what I needed to do, and she and Knox will come work on a settlement.

I still suggest that the stash can be spiked after reading the terminal, it would make better story sense that way.
 
Sorry for leaving out the details. I experienced this on the current bethesda.net "version 10" from 27.11.2020 with the Steam-Edition of the game. I dont know how this translates to the nexus-versions. I kind of didn't look a the thread title, i just stumbled here when googling the quest-bug. I didt even have a real question besides if i did something wrong to trigger this behavior.
 
Quest is still busted in 1.0.2b. I couldn't interact with the stash at all (could previously in a 1.0.0 build), because I read the terminal first? Spoke to everyone (Anton, Audra, Knox) downstairs, didn't really get any good information... the (MEDIC) option should have gotten me where I needed to go. Anyway I sided with Knox due to prior knowledge, and killed Anton. Went upstairs and now neither Audra nor Knox will speak to me.
 
Oh yeah, Anton was hostile, but then when I went up to talk to Audra, neither her nor Knox would talk to me upstairs in the separate "Hotel Room" cell.

Also, side note, what's your avatar from? Is that from Masters of the Universe?

It is Toadwart from Gummi Bears.

I would assume you tried the standard things like zone out and back in, smack them with your gun, etc.

Did you happen to anticipate Knox's script and go upstairs before he said it?
 
Huh. I never watched Gummi Bears.

I didn't try either of those things, sorry. I loaded my save, and pistol-whipped Knox, and it worked, except he remained hostile. So, fending off his attacks wasn't fun. Since he can become an enemy (if you side with Anton) maybe I should have pistol-whipped Audra instead, but I didn't want her perpetually running away (my assumption).

I of course anticipated the script as I'd done it before, but I didn't go upstairs before Knox finished telling me Audra doesn't like gunfire and to go check on her.

Yeah anticipating and acting early is really dangerous for the Fallout script engines. I learned that lesson more with Fallout 76 because quests and events could break if you skipped stages. Then it got me thinking about script breaks in Fallout 4.

Some of the S2 quests are twitchy, I have played through most of them six times now (not including reloads) and you can get an entirely different level of twitchiness out of the quests.

I had the darn quest and hated every darn second of it. We get into the Harbor Hotel and all Ashton does is walk around the damn lobby. "We better check upstairs?" Except there are no stairs. I take the elevator up and kill all of the idiots. Found a room with an arrow on the wall, but no way to get to it. Went back down to the lobby and killed Ashton and moved on to another quest. F it!

There is a hotel room with a load screen door on the upper level.
 
So this quest has been drastically improved in the 2+ years since this post was originally posted, so i doubt you, presumably on 2.2.3-something, are running into the same issue as this person on 1.0.0g.

secondly, if you are running into people "upstairs" at the top of the elevator, then it's likely you are running into a conflicting quest. i know Tales from the Commonwealth replaces some sections of this area, but i've not run into conflicts with that mod since they replace different doors, even while both quests are running concurrently.

finally, Audra's room is a small apartment behind a loading screen door in the small hallway at the top of the elevator, on the far opposite side of the hallway from the door that leads out to the rooftops. the door should be red and right next to a collapsed pile of debris blocking the hallway further into the building. you do have to go inside and discover her disgusting breakfast crimes (not really) before the quest will proceed.

Thanks. I found her chems in the apartment off to the right of the elevator, but there is no door on the far side of the rooftop. I scoured the rooftop thoroughly.

I have done all that and still when I go back to the lobby the idiot is still bitching about his sister and he is alone. I give up! It seems like I am having one of those days where Murphy is kicking my butt... (Murphy's Law.) It has been two weeks and my bruised ribs are still hurting like heck from the fall. I can barely breathe as it is. Maybe I should just not play for a while... but I am bored to death otherwise. Thanks for helping me out though. I do really appreciate it.
 
The second journal entry on the "piece of junk" terminal in her room will trigger the stand-off downstairs. make sure you look at all the journal entries.
