Probably not.

I built the tower on a layer using Workshop+, then I used Place Everywhere to turn off workshop timeout, and flew the whole thing up the hill and plopped it down.
 
I also used the W+ blueprint tool to put a duplicate tower near the water tower as a screenshot platform, but that one is not "canon" to the narrative.
 
BTW: I have discovered that Transfer Settlements handles placing objects way out of the build zone just fine.

Both Red Rocket and Sanctuary were successfully moved from one save to another.
